Enzymatic iodination of tyrosine and thyroglobulin with chloroperoxidase.

چکیده
1. Crystalline chloroperoxidase is effective in catalyzing the iodination of tyrosine or thyroglobulin when supplemented with HzOz, or with the H20z-generating system, glucoseglucose oxidase. The iodination reaction is very rapid at low concentrations of iodide (5 X 10-5 M), and at low concentrations of tyrosine (1 X 10V4 M) or thyroglobulin (0.33 mg per ml). 2. Iodide is rapidly bound as 3-iodotyrosine, 3,5-diiodotyrosine, and thyroxine during the chloroperoxidase-catalyzed iodination. After 60 min of incubation with thyroglobulin as acceptor, chromatography of a Pronase digest showed approximately 45 to 50 % of the added nlI in the form of 3,5-diiadotyrosine, about 20 to 25% as 3-iodotyrosine, and 4 to 5% as thyroxine. 3. Since iodination of thyroglobulin occurred very effectively in the presence of only a single crystalline enzyme, the results suggest that it may not be necessary to postulate the existence of a separate “tyrosine iodinase” in the thyroid. Moreover, the formation of appreciable thyroxine in the chloroperoxidase-catalyzed iodination of thyroglobulin suggests that a peroxidase may also be involved in the coupling reaction in the thyroid. 4. The chloroperoxidase-catalyzed iodination of tyrosine and thyroglobulin is greatly accelerated by 0.1 M chloride and bromide. The mechanism of this stimulatory effect is not yet known, although several possibilities are discussed. 5. Iodination of thyroglobulin and tyrosine in the presence of chloroperoxidase is readily inhibited by low concentrations of antithyroid drugs and by naturally occurring reducing agents such as cysteine, reduced glutathione, and ascorbic acid. Results obtained with these inhibitors support the view that antithyroid compounds act as competitive inhibitors in the formation of organic iodine in the thyroid. 6. Excess iodide inhibits the chloroperoxidase-catalyzed iodination of tyrosine and thyroglobulin. These results suggest that the antithyroid effect of iodide, observed under

Preferential Synthesis of Thyroxine Tyrosyl Residues in Thyroglobulin* from Early Iodinated
A double labeling procedure was employed to determine whether the tyrosine residues in thyroglobulin that are iodinated first are also those that are most readily converted to thyroxine. Goiter thyroglobulin containing less than 1 atom of iodine per molecule was iodinated first with 9 at relatively low levels of iodination (5 to 20 atoms of iodine per molecule) and afterward with lzjI at higher...
متن کاملSpatial requirement for coupling of iodotyrosine residues to form thyroid hormones.
A linear random copolymer of tyrosine and lysine and two synthetic oligopeptides containing two tyrosine residues in addition to lysine residues give thyroid hormone (thyroxine and triodothyronine) residues in good yield upon enzymatic iodination with thyroid peroxidase. These synthetic peptides may serve as simple models for thyroglobulin, the protein in which biosynthesis of the thyroid hormo...
متن کاملIodination and oxidation of thyroglobulin catalyzed by thyroid peroxidase.
The kinetics of iodination and oxidation of hog thyroglobulin were studied with purified hog thyroid peroxidase and the results were compared with the reactions of free tyrosine. From Lineweaver-Burk plots and on the basis of a value of 0.83 for delta epsilon mM at 289 nm/iodine atom incorporated, the rate constant for transfer of an assumed enzyme-bound iodinium cation to thyroglobulin was est...
متن کاملThe fate of the "lost side chain" during thyroid hormonogenesis.
Thyroglobulin labeled with L-[U-‘*C]tyrosine was prepared, purified, and iodinated in order to investigate the fate of the 3-carbon fragment which remains when 2 iodotyrosine residues (18 carbon atoms) in thyroglobulin react with each other in the course of thyroid hormonogenesis to form 1 hormone residue (15 carbon atoms). Incubation of hog thyroid slices with L-[U-‘*C]tyrosine led to the form...
متن کاملThe Properties of Thyroglobulin VIII. THE IODINATION OF THYROGLOBULIN
It has been shown that when thyroglobulin is iodinated, a marked intensification occurs in its absorption spectrum in the region of 300 to 3fXl rnp in alkaline solution (1). It is also well known that the absorption peaks of iodinated tyrosine and thyronine derivatives occur in this spectral zone (2). A procedure based on these factors has been developed which permits the simultaneous determina...
